Design Notes
The Lettuce split aptamer was developed by researchers at the Weill-Cornell Medical College using the SELEX (systematic evolution of ligands by exponential enrichment) process. For the purposes of Lambert iGEM, the sequence complementary to the left portion of the target— the complement of the middle sequence (BBa_K4245131) within the Rolling Circle Product (RCP)— was added to the 3’ end of the Lettuce left sequence.
The experimentation with this version of Lettuce left was not successful, so Lambert iGEM modified the flanking sequence to include one more nucleotide complementary to the target. The modified Lettuce left (BBa_K4245134) was more functional.
